For the treatment of breast cancer, the recommended dose is 20mg once per day to 20mg twice per day. The recommended dosage for ductal carcinoma and the prevention of breast cancer in high-risk women is 20 mg per day for up to 5 years. Mechanism of Action:
Nolvadex and its generic both contain the prodrug, tamoxifen, which is metabolized in the liver by the cytochrome P450 into active content 4-hydroxytamoxifen and N-desmethyl-4-hydroxytamoxifen. These active metabolites show 30-100 times more affinity than tamoxifen towards the estrogen receptor. Research shows that breast cancer cells require estrogen for normal growth. These active metabolites competitively inhibit and prevent the estrogen from binding to the estrogen receptors. This lack of estrogen at the cellular level blocks breast cancer cell growth by decreasing DNA synthesis.
